چکیده

• PfDHPS-A437G is known as one of sulfadoxine-resistant mutations in P. falciparum . PbDHPS-A394G berghei corresponds to PfDHPS-A437G. We generated a transgenic parasite and investigated the developments. grew mice produced oocysts with similar efficiency wild-type. The mutation may not impose fitness costs on during examined stages. Genetic changes conferring drug resistance are generally believed pathogens absence drug. However, resistant parasites against sulfadoxine/pyrimethamine has been inconclusive Plasmodium falciparum. This because conferred by complex combination dihydropteroate synthase ( dhps ) dihydrofolate reductase dhfr ), which makes it difficult separately assess extent magnitude imposed To sulfadoxine alone, we rodent malaria parasite, clone harboring an A394G (PbDHPS-A394G), corresponding causative for (PfDHPS-A437G). A four-day suppressive test confirmed that was sulfadoxine. wild-type clones showed growth rates gametocyte production. observation competitive experiments were co-infected into directly survival competition between them. In mosquitoes, there no significant differences oocyst production These results indicate alters but disadvantages blood stages formation mosquitoes. partly explain persistence mutant natural populations.

Analysis in Escherichia coli of Plasmodium falciparum dihydropteroate synthase (DHPS) alleles implicated in resistance to sulfadoxine.

Mutations in Plasmodium falciparum dihydropteroate synthase have been linked to resistance to the antimalarial drug, sulfadoxine, which competes with the dihydropteroate synthase substrate, p-aminobenzoate. In an effort to evaluate the role of these mutations in a simple model system, we have expressed six relevant alleles of the P. falciparum dihydropteroate synthase gene in Escherichia coli. ...

Genetic Variability of Plasmodium malariae dihydropteroate synthase (dhps) in Four Asian Countries

The dihydropteroate synthase (dhps) genes of 44 P. malariae strains from four Asian countries were isolated. Only a limited number of polymorphisms were observed. Comparison with homologous mutations in other Plasmodium species showed that these polymorphisms are unlikely to be associated with sulfadoxine resistance.

Dihydropteroate synthase (DHPS) gene mutation study in HIV-Infected Indian patients with Pneumocystis jirovecii pneumonia.

INTRODUCTION Pneumocystis jirovecii dihydropteroate synthase (DHPS) gene mutations' (55th and 57th codon) association with prior sulfa prophylaxis failure has been reported from both developed and developing countries. We conducted a prospective study to determine the prevalence of P. jirovecii DHPS mutations from 2006 to 2009 on P. jirovecii isolates obtained from HIV-infected patients with a ...

Allelic exchange at the endogenous genomic locus in Plasmodium falciparum proves the role of dihydropteroate synthase in sulfadoxine-resistant malaria.

We have exploited the recently developed ability to trans- fect the malaria parasite Plasmodium falciparum to investigate the role of polymorphisms in the enzyme dihydropteroate synthase (DHPS), identified in sulfadoxine-resistant field isolates. By using a truncated form of the dhps gene, specific mutations were introduced into the endogenous gene by allelic replacement such that they were und...

Tracking origins and spread of sulfadoxine-resistant Plasmodium falciparum dhps alleles in Thailand.

The emergence and spread of drug-resistant Plasmodium falciparum have been a major impediment for the control of malaria worldwide. Earlier studies have shown that similar to chloroquine (CQ) resistance, high levels of pyrimethamine resistance in P. falciparum originated independently 4 to 5 times globally, including one origin at the Thailand-Cambodia border. In this study we describe the orig...
